Best way to stop a drip campaign either 1) when a leads responds or 2) when sales person sends email manually?

The way I would do this is:

1. Remove from flow if lead clicks on a specific link or fills out a form (create an offer or CTA)

2. Sales - you can instruct sales to add them to a "remove from campaign" smart campaign in Marketo Sales Insight.

I am sure others have various suggestions!

Remove from campaign is the way to go.  Trigger the workflow step in a separate campaign

By "trigger the workflow step in separate campaign" you mean a separate smart campaign just for removal? What is the reason for this..to track who is removed better?
