Is it possible to unsubscribe contacts through Marketo API?

The unsubscribe field isn't available through the API by default but you can set up a custom field to populate via the API along with a data management campaign in Marketo or you could create a campaign with a Request Campaign (source is web service API) that unsubscribes leads and call that. I'd probably pick option a, personally.

Yes, I have seen this done as well.  You could also have an "explicit opt-in" field (or use that as this trigger field) that could be used as the trigger for your smart campaign.
